Real Life Customer Case 1:
Analysis of Antibody Fusion Proteins - Phase Screening and Method Development Service
Sample:
Recombinant tumor necrosis factor receptor-Fc fusion protein (rhTNFR-Fc), including oligomers (440 kD and 360 kD), monomer (150 kD), and impurity (120 kD)
Goal:
In search of high resolution separation between protein monomer, oligomer, and impurities.
Approach:
Screen different pore sized size exclusion stationary phases to achieve best separation
Conclusion:
Zenix SEC-300 (3 µm, 300 Å) column delivers baseline separation of Fc fusion protein, its oligomer, and impurity.
Summary:
Sepax recommends Zenix SEC-300 (3 µm, 300 Å) column for highest possible resolution based separation of protein, oligomer, and impurity peaks which later to be adopted and standardized as QC method by customer.
Real Life Customer Case 2:
Analysis of Interferon Separation - Phase Screening and Method Development Service
Sample:
Crude Interferon
Goal:
In search of most suitable stationary phase and separation conditions from analytical to process scale.
Approach:
Screen variety of stationary phases at analytical level (4.6 × 250mm) to locate specific selectivity followed by mobile phases and run conditions screens and optimization.
Conclusion:
Bio C4 (5 µm, 120 Å) column as shown on the right offers the best selectivity and resolving power among the phases screened; Separation profiles were also provided with various mobile phase conditions.
Summary:
Sepax recommends Bio C4 phase with optimized method developed at analytical scale which later allowed and assisted customer to successfully scale up to Sepax Bio-C4 (5 µm, 120 Å) and further process scale.
